I am making a werewolf comic book an would like to hear from you.

Something original is always nice.
What I look for when I buy a book or manga (I don't read American comics) are things like originality, well-drawn art, and a great story. I usually go for the type of things that are a bit suspense, gore, and dark.
But really, you can't go wrong with comics. Just sit down and write your story out first, you'll be surprised what you may add later when you're drawing.

First and foremost, I like nice art. And I agree with the above that I do not buy American comics, mostly because I feel like the art and layouts tend to be second rate, creative layouts and style can totally change how a story comes across.
However, there is, of course, much more to consider. A story-line that has real character development and which slowly reveals the histories and relationships of characters is really appreciated and one that is more about the characters striving towards some goal rather than just a pack of gore and violence is really likely to increase the quality of the plot.
Now, to answer more subjectively on my own opinions about werewolf stories in general and art based ones in particular (note: from here on out is all personal opinion with no grounds to back it up as universality):
Use of old mythology. This can be to explain it, adhere to it, or disprove it but certainly do not ignore it. if it takes place on earth then everyone should know the common myths and be prejudiced to believe them even if they are false. Furthermore, I prefer stories that either use or build on them rather than contradicting them.
Make the transformation complete physically but not mentally (actually look a wold but actually be a human for the most part)
Give the person wolfish tendencies (e.g. a growl, good insticts, ect)
If the transformation is volentary, allow to werewolf to sometimes have fangs or linger nails or pointed ears and sometimes not as moods or instincts take him.
If the transformation is tied to the moon make him wolfish as above in the days just before and just after.
Don't go for the stereotype of mopey emo tormented soul. It is overdone and I like my werewolves to have real personalities.
I am just curios to see what it would be like to have the bad guy be a good guy. Someone who honestly thinks he is doing right in hunting and killing werewolves. However, for the story to be from the werewolf's point of view (or at least sometimes from it) and the werewolf, whether he does bad things or not, to not be primarily evil.

Good story, and a GREAT transformation scene. I like the way the Howling/ Dog Soldiers werewolves look like- standing on two legs, but I think the scariest is the American Werewolf in London savage beat type.

why are the first two comments about how "I don't read American comics"? Dude, artistic style is different. Just because you like comics with big bug eyes and overly pretty men doesn't mean that it's a superior form.
Okay, now for a suggestion: As with every comic I come up to, I look for three things:
Does it grab my attention? (because some stories, no matter how original, have crappy writing and don't draw me in)
Is it appealing? (This isn't about style, it's just about the level of craftsmanship, in the writing and definitely in the art. Forget style, can you just draw?!)
Is the style unique? (This is minorly unimportant, but it is still important. Too many copy their favorite artists, or they only draw a few types of faces. First, get good at story and art, and then focus on style)
Getting those out of the way, what I wouldn't mind seeing isn't the "Imma werewolf, I'mma gonna eat you up, Imma evil" with a good hunter coming in. Nor would I want to see the "Imma werewolf, Imma innocent, Imma still human" with a hunter wrongly hunting someone down.
The distinction between right and wrong, good and evil isn't clear. A werewolf could be evil, but there could be others that are good. But what I don't like is stories making the main character and their race into the good guy, while there's another killer monster race out there.
Another thing would be to try would be either a new way to design the werewolf character (like how they did in the Harry Potter movie), or to make the werewolf mythological-ly accurate, down to the deal with the devil and the no tails thing.
But seriously, it is up to you.

Agreed American werewolf in London great one right there one of my favorite transformation from human to werewolf

Make sure that your drawings are awesome.
We tend to only buy comics for the pictures, in the end.
The story is important, of course, but do something that makes your comic worth while in an artistic sense.
